1. Currently I am running the ESI-MS under these parameter & I just want to make sure that these parameters are accurate for proper & long time functioning of ESI-MS instrument. Right Now under these parameter our ESI-MS is working fine.
LC flow rate =0.5ml/min
Heater Temperature= 300 C
Capillary Temp= 350 C
Spray voltage =5 kV
sheath gas flow rate =50 arb
Auxillary gas flow rate= 20 arb
Aweep gas flow rate=0 arb
S-Lense RF level=67

The settings of your ESI-MS depend in your analytes of interest. So there is no general answer to your question.

In addition the physical x-y position of the probe, some negative ions like the probe to be very close to the introduction system.
Helium is normally used as collision gas for MS/MS analysis, but you can use argon as well
As Dr. Schumacher points out, the optimization must be carried out for each analyte and each system. You are using a compromise set of parameters that are probably adequate for your analytes of interest on this specific system but that does not mean that these conditions will be adequate for all analytes or for other systems.
Helium is apparently your collision gas. You would be better off with argon if it is available since it is significantly more massive than helium.
