I am working with decidual cells. These cells were stained nicely with Cytokeratin7 before passage but, after passage it shows only 40 to 50% cells were stain and remaining were unstained. Is it the sign of differentiation or I need to change staining protocol?
As a startpoint, an antibody to vimentin, the mesenchyme intermediate filament protein, would be one potential marker that has been used extensively in the literature over the years? I must say that I am a bit surprised that you are using cytokeratin 7 as a marker, which would be usually used as a marker for cells of epithelial origin - that maybe why it is switched on very early, which may be inappropriate, then disappears after passage and the cells have settled down to am more usual phenotype? Thank you Simon. Actually I also use Vimentin and it shows very nice staining but another markers like IGFBP 1 and CD10 showing some confusing results. IGFBP 1 nevere stained at all where CD 10 showing partial staining (near 50-55% stained cells).

I think prolactin(PRL) and IGFBP1immunoreactivity and also receptors for Progesterone
Hello Tulay, thank you for the suggestions. I do use IGFBP1 for immunocytochemistry, but it won't work. Now a days, I am doing PCR for prolactin and it gives the results.
