What are the exact method for prepration of rabid urease test for testing for helicobacter pylori ?

i am working on helicobacter pylori project . i keep culturing it but every time it gives urease negative results

is there any other conformation method for its growth

You can test urease activity on plate by adding urease (600 mg/l final concentration) and red phenol (100 mg/l final concentration) in your Columbia Blood Agar Base supplemented with serum and antibiotics. Urease positive bacteria will increase pH. Yellow plate will turn red.
