I need to isolate these strains from ruminal fluid to use them for further investigations, but most of works published about methanogens use molecular methods. We have an anaerobic chamber in the laboratory and I found general requirement of methanogens but need more specific features of these species.
It would be difficult to isolate and characterize these two species of Methanobrevibacter as there is no specific/ different requirements for these species. But you can retrieve literature on their antibiotic sensitivity or carbon source requirement. I would rather suggest, make a request/ purchase these species from a culture collection center.
I agree with the idea of Sanjay Kumar, it is very difficult to isolate these organisms due to environmental, nutritional requirements and metabolic. However some methanogenic can be grown in tubes or bottles more easily: see if this text will help you in your search.

I agree with the idea. But then I would suggest you to grow them on a plate and make replica of these plates after you get colonies. Sent them for sequencing and see whether you got these two species or not. If yes, you can use your replica plate to get the isolated colonies and grow them for further use.
I would suggest to dilute in liquid medium. It is quite hard to cultivate methanogens on plates. It is also tricky to use anoxic chamber or a tent for this purpose. Very often cultivation inside anoxic chambers results in quick growth of inhabitants of the chambers.
