I have been using Alexa 488, 555 and 647-conjugated secondary antibodies and phalloidin, and except for the price, I’m happy with them. But as time passes I am wondering if there is anything better/cheaper around. I also would like to add a 4th channel that is excited by 405nm laser. I image sequentially using Leica SP8 confocal which has the following lines: 405, 458, 476, 488, 496, 514, 561, 594, 633.
405 AND 488, 555, 647 should go well with your microscope! The Alexa fluorhophore tagged antibodies are very good, and haven't used any alternatives... unless you buy primary that is biotynylated and use fluorescently tagged streptavidin. might be cheaper because you use it less dilute. However I would stick to the Alexa antibodies you mention.
The atto and dylight dyes perform just as well and are usually cheaper. For your phalloidin the standard fitc and tritc tagged one from sigma is much much cheaper and works great. Sure the flurophores are old school but there is so much actin in a cell it doesn't matter
