I would like to suggest you that you should culture some good tumor cell line in your scaffold as well as in a conventional plate as a control. After enough growth or spheroid formation you can isolate and inject to mouse at the same amount. For detail injection you can use any reference paper which is easy for you . Both of your injected cells will make tumor in mouse model and then you can compare time dependently the size as well some most commonly used markers expression level by RT-PCR or western. If your constructed scaffold provide a more valid micro-environment than conventional palte culture then you can clearly proof by using above experiment.
