Safety Skills during laboratory practice is the basic thing which must be follow prior to work ... not only in microbiology field but also in other fields of science and engineering.
In the field of Microbiology, one should know about the safety regarding chemicals, dehydrated growth mediums and sterilization techniques (sterilization of labwares, preparation of chemicals, sterilization of growth mediums, laminar flow cabinet, and the culture room conditions). If one does not know about the sterilization techniques then handling the microbes and their culturing is difficult and may prove dangerous (if working with pathogens).
Yes i agree with Kunal its important to posses basic lab. safety skills!
Also things about sterilization of labwares, mediums ect is MUST before commencing experiments.
You need to read or better to see someone skilled doing the cultivation, spreading the cultures on plates, how to hold and use the instruments during the experiments.
A great caution is needed while handling with pathogen strains!
Those are some basic stuff that i think are important to know before you start working on your own.
I agree with Kunai and Nicola but all of these must be hands-on under direct supervision of a microbiologist.. Reading and observing look easy but will not help you get the skill in performing the tasks with the hands. Make aseptic techniques your by-word and post notes in your working area (laminar flow,isolation area and other working areas) to remind you. In due time you will have the dexterity to start a microbiology work, along with patience and perseverance.
