What are possible reasons for failure of separation of nanoparticles by centrifugation?

I centrifuged a 14 ml sample of chitosan NPs of 60 nm diameter for 1 hour at 20,000 rpm and the solution remained clear as it is.

It remained clear because maybe chitosan molecules are still in solvated state. You need to increase the solution pH value in order to separete chitosan.
