I`m about to start my thesis on breast cancer immunotherapy which is designated to examine a nano particle-based vaccine for the delivery of tumor associated antigens to DCs in vivo.
However, I need an effective immunostimulatory adjuvant besides the vaccine in order to elicit a better effect, since combination therapy has been proven to be more effective.
I need an adjuvant that in addition to its immunostimulatory effect, it could have the potential to be used in clinical trials..
Some preclinical adjuvants with promising results are Cpg oligodeoxynucleotides or CpG ODN. These are short synthetic sequences are TLR agonists that stimulate cytokine secretion and antigen presentation by macrophages and DCs leading to an enhanced anti tumor immune response.
NanoBio has a nanoemulsion adjuvant that can be delivered either intranasally or intramuscularly. When administered intranasally it elicits a balanced Th1/Th2 systemic immune response as well as a mucosal immune response. The nanoemulsion can be mixed with recombinant proteins or live organisms resulting in lysis of the pathogen and incorporation of immune determinants into the nanoemulsion droplet which are then delivered effectively to dendritic cells by either route. The adjuvant has been evaluated in two Phase 1 clinical studies with flu antigen. It has been shown to be protective in numerous challenge models in animals including RSV and HSV-2. The nanoemulsion has undergone preliminary testing in mice in a breast cancer metastatic model with promising results.
