To examine a kind of cytokine protein level change of a representative area in somatosensory cortex in brain tissue followed by peripheral nerve injury, may I know what kind of biochemical technique is best to exam? ELISA, Tissue array, Tissue Microarray,  IHC or Western Blot?

Previously I planned to do ELISA, but I have challenges on how to precisely get the tissue out from the specific area. Besides, my colleague challenged me that ELISA is mostly for supernatant, it's not appropriate to apply on my study. He suggest Tissue array may be more appropriate to address my concern.

However, I did found evidence of checking protein level of brain tissue by ELISA, I don't know how to justify my doings.
