PCR is used in the early stages of processing DNA for sequencing for detecting the presence or absence of a gene.
Main principles of PCR:
1) DNA denaturing
2) DNA annealing
3) DNA extending.
PCR technique:
1) DNA template
2) Primers
3) dNTPs
4) Taq polymerase
5) Buffer
All are taken in specific amount in PCR tube and set up the temperature, time, cycle etc. then run the PCR machine.
