I have been attempting to cultivate E.coli biofilms using 96 and 24 well flat bottom plates, but most literature I found suggest conical bottoms. I am looking for the best 96 well plates to purchase but I'm lost on fishers site. Please let me know what 96 or 24 well plates have worked best for you when growing E.coli biofilms.

I've had success growing the biofilm with the flat bottom plates, but they tend to only grow on the surface.I'm trying to primarily grow them on the bottom of the plates so I can wash and stain them. Based on the absorbency rates I've been getting after staining and destaining most of the bacteria is being washed out.
I've been culturing the bacteria using the a basic M9 glucose but at 37°C. I can try lowering the temperature and seeing if I can get any results. I'm also culturing S.aureus simultaneously, but I've had great success with S.aureus.

How long are you typically incubating your plates? I usually leave them for about 24 hours. What do you recomend?
