I have performed many wet and semi-wet western blot protein transfers, and I have never seen something like that. Perhaps, in my experience, you should confirm the sandwich before you start the wet transfer. I remember that some time ago I had a problem similar to yours and I solved it by checking the transfer equipment available in the lab and found that the system clip didn't work so well so we had to change it.

Hi Felipe Arroyave thank you for your tip, I will check the equipment. The strange thing here is sometimes I see the problem, sometimes no. Even when I transfer 2 membranes at the same time, it happens at different extension in both of them and usually at the center part of the membrane, although it happened also at the ladder region. I will check the equipment. Thanks.
Hi Yssel, this is the typical pattern that you obtain when there are bubbles trapped in your sandwich between the gel and the membrane.
