Hi,

I'm fairly new to western blotting but have had reasonable success up until now. Despite being able to successfully detect and image my target protein (p-ERK), I'm having real issues with my housekeepers. I can't seem to detect α-tubulin across samples (have tried multiple primary/secondary Abs), whereas β-actin is occasionally detectable but does not correlate with target protein (e.g. lanes displaying high p-ERK expression have no β-actin present, whilst lanes expressing no p-ERK do have β-actin). Revert total protein stain also shows no protein. Very confused!
