We are currently working on setting up an assay for detection of Vibrio using filtration and culturing on TCBS-agar. The culture grows without problems on the TCBS-agar when plated directly, but when we try to spike a PBS-3% solution with bacteria and then run it through a 0.22uM MCE membrane (EZ-FIT Filtration Units) and filter it using a EZ-Vakuum pump before placing the filters on the TCBS-agar, no growth is found on the cultured filters. We have run Enterococcus through the same protocol and get large amounts of growth on the filters then. Any suggestions to why the Vibrio is not compatible with the filtration-step? All help is kindly welcomed!
