I am working at the plant immunity lab and I am working on two isolates of Botrytis cinerea. The media I used was 4% agar and PDA and incubate them in 24 hr light and one isolate growed well but the other isolate has some problems. I didn't see any spores and only hyphae was occurred and could you please suggest me the growth conditions for sporulation of this isolates??? Thanks in advance.
I think PDA is good for its growth. But for sporulation, you can also keep under refrigerated condition under 4degree celcius just like that of Pyricularia
The optimum temperature for spore germination is 18–21°C. Some spores still germinate at temperatures below 10°C. Light treatment can also effect the sporulation. PDA is good for growth.
1. V8 juice ( 100%, original vegetable juice )
2. Bacto-Agar
3. Calcium carbonate
4. Mycological peptone
5. Sodium phosphate
6. Maltose
I grew fungi on V8 media at 25*C and dark conditions, the spores inoculation was occurred. Thanks a lot for all of your answers.
