I collected root samples from different tree species at different sites in different seasons. Every time I processed the samples until the step before DNA extraction and stored them. Samples have accumulated and I have now extract DNA from them. I did DNA extraction from 60 samples at once last years and I could get good results. I would like to know what is the maximal sample size that only one operator can handle in such an experiment without failure.
A. Kevin G. Djotan, are you using spin column tube format or 96 well plate format? If you are using spin column-based protocol then I would suggest going for 20 to 30 samples at once (depending on your confidence) considering a basic micro-centrifuge rotor can hold up to 24 or 30 tubes at once. On the other hand, if you are using a plate-based method then you can process at least 2 plates (192 samples) at a time. In both cases, you can finish the procedure in about 2 hours.

Pushkal Sinduvadi Ramesh , thank you for the recommendations. Yes, I am using the spin column-based protocol. Considering the micro-centrifuge rotor capacity, 30 samples at once as you suggested sound a good idea. I remember that due to centrifugations, it was tough when I used 60 samples at once.
