When interaction is significant in the 2-way ANOVA test, it makes no sense to interpret the main effects of each factor. In this case it is necessary to unfold the interaction. This is done by fixing one of the factors and at each level of the fixed factor, you compare the levels of the other factor with pairwise comparison (Example: Tukey). The link in Usman Rashid answer will be very usefull for you.

However, please, I am still not comprehending very well what to take as replication effect or triplicate effect or interaction. I have actually ran the Tukey test but I Don't know where to read the replication or triplicate interaction. Could it be the LS mean??

Please, can you explain more on how to fix and unfold interaction because it is not explain on the link.

In the two way ANOVA (location by genotype), what is your outcome variable?

I don't think that replication and triplication effects are statistical tests in this context. Did you study replication and triplication as part of your study?

Difo Voukang Harouna You already have most of the results you need. There are differences across species. This is also clear from the means presented in the first three rows. I think what the reviewer is asking is: Are there any differences across species within location A and location B? It seems like there are.

This can be answered by performing pair-wise comparisons using Tukey's HSD.

The results will look like following:

Location = A:

Specie 1 - Specie 2 =

Specie 2 - Specie 3 = ...and more

Location = B:

Specie 1 - Specie 2 =

Specie 2 - Specie 3 = ...and more

Did you perform pair-wise comparisons?

I think you have solved my worry if that is the case. However, please can you explain to me briefly which part could explain the triplicate effect there. I also see that it is comparing between locations A Vs B (contrast) but i found this other attached may be it is the right one, could you advise.

Please, one final issue I kindly wish you to clarify me; As you mentionned th there are some interactions within location B only but could that mean the réplications were wrongly done or have any implication on the interpretation of the réplications.

The attached image is of model parameters. Presenting this may also be helpful but this is not what the reviewer is asking. The reviewer is asking for differences between pairs of species within location A and differences between pairs of species within location B.

You have not done anything wrong. All you need to do is to provide the output of pair-wise comparisons from XLSTAT.

For example: At cite A. The species differences are:

Specie 1 - Specie 2 = 10.24 - 16.29 = -6. If you get this from XLSTAT, it will also give you standard error, confidence intervals and z-values and p-values.
