If I cultured pluripotent cells on ultra-low attachment plate in order to form Embryoid Bodies, and after 2 weeks I isolated the RNA from these cells and I found that the differentiation gene expression decreased significantly. What can you conclude?

1- Failure to form EB. (Not optimum protocol)

2- Can ultra-low attachment plate be considered as a selection criteria to select only pluripotent cells while other less pluripotent die as they need attachment.
