Yes. Take a look on literaturw. Your concept is ok, but remember that you need to "inject" continously dractions from 1st D to 2nd D, thus the valve between the columns must have a loop (s) (read about modulation/modulators dor 2D LC)

"Valve" between two columns does not need any loop. No need to inject continuous fractions unless application calls for it. Depends on what the goal and need is.

Once the primary methods have been optimized for use, most 2D HPLC methods are fully automated. *We have developed many custom 2D LC methods for major pharma companies over the past decades which were used for both initial analytical scale methods as well as in preparative scale-up methods for production.

You certainly can "build" your own 2D HPLC system yourself, but a detailed understanding of the flow path, the delay volumes, column volumes, application(s) and proposed methods are all required to make it work well (easier said than done). Best for highly skilled users with lots of method develop experience. This is a case where even having the best "tool" for the job is not enough because developing the best method to take advantage of it is the hardest part. In general, never use 2D HPLC to solve any problem, unless you have no other options. Practical to design your own system? Probably not.
