I recently used MCE's TO-PRO 1 (HY-D0917, MCE Product) to conduct nucleic acid staining experiments. I would like to ask about the optimal staining concentration of this reagent.

Regarding the optimization of the staining concentration of TO-PRO 1 (Cat.No.: HY-D0917), the following parameters can be referred to according to the differences in the detection platform:
1. Flow cytometry detection
The recommended working concentration is 25 nM~1 µM, which is suitable for nucleic acid staining of suspended cells (such as cell cycle analysis). Note:
-Low concentration (25~100 nM) is suitable for samples with high nucleic acid content (such as G2/M phase cells);
-High concentration (500 nM~1 µM) is recommended for nucleic acid degradation samples or low-abundance nucleic acid detection.
2. Fluorescence microscopy detection
The conventional working concentration is 1~10 µM, which is suitable for:
-Nuclear staining of adherent cell slides (5 µM is recommended);
-DNA counterstaining of tissue sections (8~10 µM is recommended for frozen sections to enhance the signal).
