Has anyone tried to transfect EBV-transformed B-cells with XtremeGene transfection reagens (produced by Roche)? (Would you recommend a variant "9" or "HP"?)
In fact, I did not try any at the moment, just planning.
My idea is to start with the XtremeGene HP, as this worked better with the cells growing in suspension. I've tried to find some recommendations about transfection of EBV-B cells, but there was just electroporation method mentioned. Not our option.
We had some successful transfections using XtremeGene HP reagens (Roche). We used 600 000 cells in 1 ml of RP-MI + 10 % FCS, and transfection mix of 50 ul RPMI, 1,2 ul tr. reagens (XP) and 400 ng of DNA. We did not measure the transfection efficiency, but got a good quantity of specific transcript. Hopefully this helps.
