My pictures are in jpg format, I need to know if it is possible to quantify fluorescence of S100B and GFAP in cells. And wich is the best way to do this, using mean gray value or integrated density? In both cases I do not have clear how the measure is, wich is the units or how can I know this? I attach a sample image for a marker, Thanks!!

I measured your image. For 9 cells, integrated/area gave me an RFU of 9.6579 +/- 2.6793, while mean gave me 9.6578 +/- 2.6794. Integrated gave me 559394.1 +/- 316727.2. This was with the image scaled to 32-bit.

While agreeing with the necessity of non-lossy image formats for analyzing images, I would like to add that it may be very useful to have the BioFormats plugin installed in imageJ. You can do it yourself by manually installing it from
https://imagej.net/Bio-Formats
or instead of ImageJ itself, you can install Fiji, which is a plugin-enriched version of ImageJ available from
https://fiji.sc/
The advantage of the BioFormats plugin that it can import most microscopy image formats (Zeiss CZI and LSM images, Olympus OIF, etc).
