I have alloxan monohydrate and Sprague dawley rats in my lab. I dont have Streptozotocin. Right now, I would like to study DPP IV inhibitory activity of my compound in in vivo model. So, I have doubt that shall I use alloxan to enhance DPP IV expression? Is it correct way ?
Two things can increase the expression of DPP IV. One is insulin deficiency and the other is high fat diet. you can induce insulin deficiency by any of the two compounds i.e alloxan or a low dose streptozotocin combined with high fat diet.

I agree with the suggestion of Dr. Jehan Zeb Khan in this regard. However, personally I would prefer using Streptozotocin for inducing diabetes as this would be less toxic.
To obtain a lower death percentage in rats following the i.p. injection of anti pancreatic beta-cells, my experience show that alloxan is only indicated to rats above approx. 250 g bodyweight, while STZ both to rats above and below that bodyweight. The results - diabetes induction - are identical. So, I definitely recommend STZ combined with food depleted of nicotinamide. To only induce diabetes, the rats don't need a fatty diet.
Thank you for your kind reply sir. Actually I have purchased alloxan for my experiments based on literature survey. Right now I am unable to purchase streptozotocin again for same purposes. Shall I use ALLOXAN only for enhance DPP IV expression in my studies?
