Has anyone got any advice on how to reconstitute lyophilized material, contained in a 3mL glass vial, through an igloo cap, with only 125uL dilution buffer? I find that the volume is so small that it cannot overcome its own superficial tension and viscosity, so it is really hard for me to rotate the vial and ensure that all the internal surfaces have been reached. In fact, even spending a long time rotating the vial, I have no guarantee that I managed to recover all the powder. Unfortunately changing the vial size or diluent volume is not an option, as it would require full requalification of the manufacturing process. Any technical tips?
