Hi,

I am new with Western Blot and I need a few clarifications about the use of blocking and incubation solution. I understood that blocking is done to saturate the membrane with proteins non-reacting with the antibody so that this latter can bind the specific protein..But in practice, does it mean that the antibody compete with the proteins in the blocking solution for the binding to the protein on the membrane?I mean, the protein of interest( that is present on the membrane after the transfer)will also bind the proteins in the blocking solution before the incubation with the antibody?And then, during the incubation with the antibody, this latter replaces the proteins of the blocking solution in the binding to the protein on the membrane?

Also, why also the ladder gives signal after finishing the western blot and visualizing it?
