The mobile phase is Toluene:Ethyl acetate:Formic acid: Gallic acid (4:5:1:1) reutin is showing two spots and in methanol it is not showing two spots.

If the standard forms two spots means it forms two solute fronts one near the baseline must be due to impurity and the other spot in top is attributed to parent standard quercitin.

It might be due to slight impurities. Did those spots have the same colour under UV lamp? Quercetin is UV active.

You may also do a double migration to see the behavior on your TLC card.
