Hi all. i am studying the anticancer activity of natural pure compound, the IC50 of it is 25µg/mL and the positive control that i was used is 2µg/mL, i was go deep with the compound and i investigated itʼs ability to induce apoptosis, the results showed it induce the apoptosis through intrinsic apoptosis pathway.
So how i can justify that, natural compound can be considered as anticancer even it has low toxicity

First of all, which type of model you are using? Is it a mouse model or cell line?
If it is a cell line then, I have a suggestion for your experiments.I am also using such natural compounds to check the anticancer activity in my research work. In literature, you will get the IC50 value, but for your particular cell line, you have to check the IC50 value.
So you have to standardise the dose of your drug. First, start with very low to the high dose of your drug, may be ranging from 1ug/ml to 100ug/ml. You will get a dose dependent cell death rate. Suppose you got that in 1-10 ug /ml you are getting the maximum cell killing effect. Then you set up another experiment where you use a drug dose ranging from 1 to 10ug/ml concentration, like 1,1.5,2,2.5,5,...10ug/ml.
So now you will get the exact IC50 value of the drug on your specific cell line. I got different IC 50 values of drugs in different cell lines.
And please use a control cell line, where your drug will not show any killing effect in low or high concentration. If it is not showing a killing effect on the control non-cancerous cell line, then you can justify your drug as an anti cancer drug with low toxicity in low concentration.

for my compound the acute toxicity study and in vitro cytotoxicity on liver cells also was done it is safe. but the main problem is the standard range of IC50 of the natural anticancer compound is so narrow less than 20 microG/ml in some referances. so how i can introduce my compound with good activity (apoptosis inducer) but it has high ic50.
do you have nay referances or articles on the same trake ?
