I tried several mounting alternatives to observe bacterial cells by fluorescence microscopy but none of them made me happy enough. Now, we are just embedding glass slides with 1% agarose in PBS (95ºC) and then tilting the slide to allow agarose excess fall out, so a fine agarose layer is formed. Then we just put 10 ul of bacterial suspension and over him a cover slip.

The good things of this method are that agarose do not interfere with fluorescence signal and is cheap.

The bad things are that often agarose layer surface is quite irregular, leading to different optimal focus in the same frame and the lack of anti-fading agents. Additionally, some bacterial cells doesn't fix to the surface and keep moving along the frame.

It would be nice to improve our mounting method... Please could you share your experience about mounting bacteria?
