The C57BL6J mice lack the Nicotinamide nucleotide transhydrogenase (Nnt) gene. The Nnt gene is important for glucose homeostasis and control insulin secretion. I may advice you to use another C57BL6 background for instance C57BL6N.
We usually use 4-week-old C56BL/6J mice and feed them high fat diet (60% calories from fat, prepared by Research Diet Inc.) for 12 weeks. The mice then become obese and develop hyperglycemia and insulin resistance.
