The activity of a kinase is 16 nmol/min/mg, how to calculate the amount of substrate protein and kinase, and the total volume of reaction solution?

Hi,

I am doing in vitro reaction with a kinase and its potential substrate protein. We got recombinant kinase and substrate protein. And we know activity of the recombinant kinase is 16 nmol/min/mg. How to calculate the amount of kinase and substrate, and the total volume of my reaction solution I should use?
