The events occurring during Epithelial mesenchymal transition (EMT) include the loss of adherents junctions and the down-regulation of cytokeratins and E-cadherin, epithelial specific markers, and by the increase of mesenchymal markers, such as fibronectin, N-cadherin, and vimentin, the gaining of a fibroblastoid invasive phenotype.

It is know that TGF-B1 induce EMT by down-regulating E-Cadherin and Unregulated N-Cadherin.

In my experiment, I treated Calu-3, lung adenocarcinoma cell line, with 10 ng/ml of TGF-B1 for 48 and 72 hours. As shown in western blot, the treated cells become more elongated and get mesenchymal shape, while the unexpected results was in Calu-Ctrl, N-cadherin was up-regulated and and Cells treated with TGF-B1, N-cadherin was down-regulated.
