I've read some literature that used saline as a sham injection for the control rats, and some used citrate buffer (vehicle) as the sham injection.
Furthermore, the studies that used saline has the sham injection gave the rats a relatively higher volume compare to the citrate buffer injections.
Does anyone have any insight on if these two make a difference?
Both, normal saline and citrate buffer shall not make any differnce. As you have quoted that some studies studies that used saline has the sham injection gave the rats a relatively higher volume compare to the citrate buffer injections. It can not be if NORMAL SALINE (0.9%) is used.
In case STZ is used there's no much difference, citrate is preferable for alloxan.
We use 0.1 mol/l citrate buffer, pH 4.5, to dissolve STZ. The control animals receive an equivalent volume of the above-mentioned citrate buffer.
Citrate buffer as mentioned above. But if you used nicotinamide as a protective agent before STZ inlection, then normal saline for your controls 15 min before citrate buffer injections
I am agree with Peter Owira and Omotayo Erejuwa .you can try by using citrate buffer by maintaining the PH level .
Use 0.1 M citrate buffer at PH 4.5 for prolonged stability of STZ.
I had previously used saline and my result was okay. However, on a recent work I got a different brand of stz called streptozomacin and I lost all my rats within 20mins of injection. I am suspecting stz brand used. Can some help?
