I am going to study the effect of a mast cell stabilizer on histamine content in rat kidney tissue. What are the methods used for this purpose .. chemical methods, colorimetric ,ELIZA , .... etc. Many thanks

Hi. There are some commercial kit like ENZO which measures histamine from tissue, serum or urine. The method used is ELISA. Histamine in serum usually disappears within 20 minutes after a challenge but you may also try measuring Histamine metabolites in the urine such as the N-methylhistamine (NMH). You may also correlate that with serum tryptase level which are released from mast cells. Since mast cells participate in inflammatory response and conditions such as diabetes and obesity, tryptase level in these conditions seems to be elevated. So, another area to cover for your STZ induced diabetic mice could be the Tryptase level besides Histamine. Just a a suggestion. Thanks.
