Hello. I want to ask a question about microbial identification. In our lab we stained the strains to see that is it rod or cocci. After that we sent it somewhere to analyze and identify strains with PCR. Results came bu we confused about it. Because the identifications are contradictory. For example we saw rods but it came coccus genus. We didn't understand until now. Thank you for your help...
First of all check all your steps and agents.
It's hard to answer your question because there is no details and there is many factors involved.
In other side it happens some times, just last week, I stained a pure colony, under microscopic it was diphtheroid form but after digging turn out it is staphylococcus aureus.

It could be as simple as a crossover contamination when you did the PCR-Sequencing reaction. Its hard to say with the provided information. Did you used boiling method or a standard DNA extraction?
I don’t know Ali Javadmanesh. They performed before I came to that lab. And unfortunately no one knows how hes done. He did not send the seq results either.
