Fortunately I've had some very useful advice thus far from my professor, and I'd be asking him again for more advice in this case if I wasn't too pressed for time to await his response. I'm trying to assess the kinetics of bile acids binding to aptamers (biotinylated aptamer as immobilised ligand, bile acid as analyte, using a Series S Sensor Chip SA in the Biacore T200 using Fc1 and 2 only) in PBS with 0.01% tween-20 and the other necessary salts for folding in the immobilisation buffer. All of this is fine, the only thing I am really unsure of is what solvent to use to solubilse the bile acids. Trying to avoid using DMSO. Would it be alright to use methanol or would that influence the readout?
