We have some plant product partially purified powdered form,i need to check anticancer activitiy of that.iam dissolving product in Methanol and further dissolving in growth medium for MTT assays.its showing anticancer activity.my question is Is methanol ok? or we need to do in DMSO or some other solvent,because many papers say DMSO.
In our lab, the first experiment is always checking the solvent effect in cell culture. And also, for each assay, beside the samples and the positive control, we use solvent control.
Try it with methanol, if it is the best solvent for your sample.
In our experiment we have been used for a long time, a mixture of ethanol: DMSO (2:3). We found it can solve almost all samples from plant. Even though, we always include a control for each assay.
you can use some solvent as methanol,ethanol,DMSO, chloroform... but, you must do on your plate that must have include control (only cell and medium), possitive control (cell+medium+solvent) and samples (cell+medium+solvent+product)... the solvent control is done according to ultimate dose, and the solvent amount should not effect cells. For example, you can use 5 µl/1000 µl maximal...

I checked the methanol+medium+cell control there is no effect on cell viability.
