Hi,

To determine polysaccharide composition of the cultured tobacco cell walls, I follow the protocol below.  http://www.nature.com/nprot/journal/v7/n9/full/nprot.2012.081.html 

However, the thing is the resulted cell walls are not round and clear as cell walls. They probably can be cell wall materials.My question is that are they OK for polysaccharide determination? If not, how I can improve the quality of them and obtain the suited walls for further analysis?
