I am confused as to whether is it necessary to conduct in vitro antiviral study in 3T3 cells, since I'll be conducting antiviral study on balb/c later. My compound has proven its antiviral activity, along with suggested mechanisms using vero cells. My supervisor advised me to screen again the antiviral on 3T3, in case there's any antiviral masking by the formulation vehicle (the compound will be in topical form). My problem is I can't find correlation study on in vitro/in vivo.

For skin irritation, I have conducted in vitro cytotoxicity in vero cells. But I don't know how to do correlation with my current in vivo data. I can only find skin irritation in vitro assay using 3D cell culture or reconstructed human keratinocytes. These are not applicable in my lab. Another option is to use a 3T3 NRU cytotoxicity assay and conduct registry of cytoxicity prediction model (King & Jones 2003. In-house assessment of modified cytotoxicity assay. Toxicology letters.) This is possible to do if I can get 10-20 reference chemicals suggested in that paper.
