I have problem in germinating elephant apple seed because of hard seed cover. Please suggest me how can i germinate it? Seeds are mucilaginous in nature and have hair on its surface.
Bir Bahadur Anwar Shahzad Andrew Paul McKenzie Pegman thanks for your suggestions sir... I did these techniques earlier but don't get results so i ask this question. If anyone have any other answer, please reply.
Please use acid treatment 2-5% H2SO4 in water (Soak seeds for 6-24 hours), it will remove mucilage and soften seed coats. > Gibberelic acid treatment about 10 mg/l in water for 6 hours > Transfer to mixture of soil and sand or you can use vermi-composte or perlite for seed germination. Within 7-10 days seeds will shows first sign of germination. Please note that prior to this experiment make sure that you have collected seeds from fully mature and ripened fruit. Viability testing: 1. When the seed coat is impermeable to water or when the embryo requires after-ripening, the embryo must be excised. This procedure involves removing fruit parts and the seed coats. Good sanitary conditions should be maintained, since the exposed embryo is very susceptible to microbial attack. Seeds are viable if the cotyledons turn green and spread apart. 2. > Remove the seed coats from six seeds of your assigned seed group. > Cut seeds in half and place in container of TTC. > Container will be incubated in warm (30°C [86°F]) water for 1 hour. > After incubation period, decant off the liquid. > Rinse seed halves with distilled water until water is clear. > Blot seeds on dry towel; don’t crush seeds. > Observe color. Actively respiring tissue will stain red.
