While doing the Sandwich ELISA, in general, the capture antibody was added to the wells and kept overnight at RT. For a longer period, you can coat the wells with capture antibodies, seal the plates, and store them at 4°C for further use.

You can simply store a coated dry plate at 4C for some time, but consider, before drying, blocking and stabilizing with an appropriate protein and sucrose or trehalose, or a commercial product sold for this purpose, depending on your budget.

Greiner plate guy myself, aside from that I'd start with super clean water and buffer chemicals, because anything can and will out-bind your antibody at such low coating concentrations. Consider buying coating buffer so there's someone to blame!

Many details go into optimizing and hardening an assay. It is my bread-and-butter.

Thank you very much for your suggestion, today i'm going to test the plate that i stored last week with a "simple" storage buffer (PBS1X; Glycerol 10%; Sodium Azide 0,02%) and hope for the best. For the storage buffer composition, i considered what we already use to store affinity chromatography resins. I will definitly also try the conditions that you suggested.

Another question: i plan to test different detection antibodies for the protein I'm working with. Specifically, each detection antibody recognises a different domain of the protein. As the ELISA assay configuration will always be the sandwich one, is possible to derivatize the plate with capture antibody, perform the blocking and also incubate the plate with my target protein and store it for later use when I have all the detection antibody i have to test? In this condition you suggest to use a storage buffer or dry the plate as it is? Thank you in advance for your answer
