For my research it's needed to measure the concentration of the following sugars present in a potato tuber: sucrose, fructose and glucose. Is there a reliable method to measure these with a refractometer? What is the sample preparation?

Refractrometer will measure only total soluble solids....you have to go for separate analysis of glucose , fructose and sucrose .....
Dear Anoop Kumar Srivastava thank you for your response! If it's a hard process to purify my sample and only measure these three sugars (can be in total) I will find other means that can deal with the matrix.
Hi! Have you considered HPLC analysis?? You can check this method to quantify sucrose, fructose and glucose in potato tubers with a RI (refraction index) detector
