Hello Poornima. As Ali mentioned above, there are many protocols available. You could try pulverizing your tissue under nitrogen, extracting with a lysis buffer, clearing by centrifugation, precipitating with TCA/Acetone, removal of TCA with acetone wash, and suspending pellet in IEF loading buffer. You could also try treating your tissue chips with cold TCA/acetone, washing 2-3 times with cold acetone, and then homogenizing the tissue remnant directly in IEF loading buffer. This latter approach would require small (ca 100-200 ul) volume glass-glass tissue grinders.
