Hi all. I am wondering if muscle striations can be disrupted due to poor sample handling? e.g. fixed tissues dissolved in sucrose over extended periods of time or natural deterioration or from contamination. Thank you.
Usually, a pinch with a forceps could induce a deformation of muscle fibers. In frozen muscle tissue, a previous step in sucrose bath or fixative bath, muscle fibers usually can shrink. The best way to study skeletal muscle is to freeze it in an isopentane bath cooled in liquid nitrogen as described in "Muscle Biopsy: a pratical approach" written by Victor Dubowitz.
