RL95-2 Proliferation problem in alginate matrix do you have any experience?

Im working with RL95-2 AND THESCs cell line's and have the same problem with culturing them in alginate matrix (3.5%), from day 1 they start fading. I test viabillity using Celltiter glo (promega).

When culturing in flasks i have no problem so i think i dont have any problem with cell media or supplements.

would appreciate any idea,
