Hi, i am doing tissue culture works on Xylocarpus granatum, Xylopcarpus mekongensis and
Exoecaria agallocha. Especially these plants are true mangroves . Hence i am having difficulties on regeneration in this process. so kindly give any suggessions, idea or reprints.

Plz u use the somatic embryogenesis pathway. U must try these steps, it will be helpful for u.
1- Use mangroves root explant for inducing SE (somatic embryos).
steps:
Transfer the explants in liquid MS medium containing 2,4-D 1, 2 , 3, 4 and 5 mg/l. and transfer to shaker for 12, 24 and 48 hrs.
After thats transfer the explants on sterile filter paper and blotted dry.
Transfer these explants on MS semisolid medium containing Just half concentrationof 2,4-D (viz-0.5, 1.0, ..........) fortified with NaCl 100 mg/l+ Glutamine 400mg/l+60 g/l sucrose.
U can aslo repeate same experiment with B5 A stock+MS B+C+D along with 2,4-D.
