I have two questions. I want to analyze FSH, LH and estradiol 17β in cattle during the estrus period using RIA or ELISA techniques. Please let me know, how much interval is required between these three hormones. Estrus period in cattle is about 18 hour or 24 hours. Somewhere, I have read 2 hours, 6 hours and 8 hours So, I am a bit confused.
Other question, if I will use 100 animals, so randomly how many animals I should use for the above hormonal analysis. Need your help, please.

100 animals are enough, whatever time intervals you decide after literature search , keep the blood collection time of the day as fix time.
Your project is very laborious work, I will recommend to go thru basic estrus cycle and hormonal profile literature in great detail and based on that you decide the frequency of bleeding, and store serum or plasma in aliquotes at right temperature for future analysis. I have done these kind of project in cattle for progesterone RIA and briefly estrogen also.
