For calculating entrapment efficiency using UV-Vis, the unencapsulated drug is discarded with supernatant so that you measure unencapsulated drug in supernatant via UV-vis absorption.However, the drug is insoluble in water and the removal of the drug is possible only when it is dispersed in water or dissolved in water. I want to ask how will we able to remove unencapsulated drug if it cant be dissolved in water?
If the encapsulating particles are suspended in an aqueous medium in which the drug is insoluble, the drug should precipitate. A gentle centrifugation (not so fast as to pellet the particles), or even no centrifugation, should bring the precipitated drug to the bottom of the container. The particles can then be separated from the precipitate.
