thanks.you mean i have to dilute my cDNA.as i told you that i got smear in both 3' and 5' Race in first round of PCR and i tried both touch down and normal nested PCR program.but same result is there. i have been trying 2.5mM dNTP 4ul and 8ul respectivley.please also tell me about the concetration of primers also.

if any suggestion regrading this will also ne appriciated thanks
