A quick series of questions about packaging rAAV:
I want to produce AAV vectors, however I'm having difficulty finding the helper plasmids to package them. Any ideas where to look (I haven't had much luck on Addgene)?
Also, I'm aware that you can either use a 2 plasmid packaging system (pRepCap + pAdeno genes) or a single plasmid system. Is one preferable to the other?
And finally, eventually I'd like to go in vivo with these vectors targeting the pancreas. My understanding is that AAV-6 and AAV-8 are best suited for this (or perhaps AAV-DJ/6 or AAV-DJ/8). Does anyone have any insight into this?

Thanks for your message, it's very helpful.
The model I'm wanting to use is a Kras/p53 driven adenocarcinoma model, and the cells I'm wanting to target are most probably the acinar cells (however if other cell types are hit then that is also fine).
For the route of administration, I was going to use a high titre i.p. injection, however I hadn't ruled out the possibility of doing i.v. delivery (tail vein). After reading your article however, I notice that AAV9 did not work via the i.v. route.
Was there a reason you went with AAV9 and not AAV6 or AAV8?
Also, you wouldn't be able to tell me where you got your packaging plasmids from would you?
